摘  要:目的 :研究吗啡依赖对小鼠腹腔巨噬细胞主要组织相容复合体 (MHC )和协同刺激分子 CD80表达的影响。 方法 :BAL B/ c小鼠随机分为对照组、吗啡依赖组、吗啡戒断组。吗啡依赖组采用后肢皮下注射剂量递增的吗啡建立模型 ;戒断组在注射吗啡前 30 min皮下预先注射 1 / 1 0吗啡剂量的纳洛酮 ;对照组给予相同体积的生理盐水。每组 1 5只动物 ,各组又分为 3个亚组 ,分别连续皮下给药 3、5、7d,并于末次给药后处死动物 ,分离获得腹腔巨噬细胞 ,利用流式细胞术检测巨噬细胞MHC 和 CD80分子的表达。结果 :在连续给药 3d后 ,吗啡依赖组和戒断组小鼠腹腔巨噬细胞 MHC 和 CD80表达量较对照组均无明显变化 ;连续给药 5 d后 ,两组 MHC 的表达较对照组分别下降 4 0 .4 %和 2 8.0 % (P<0 .0 5 ) ,而 CD80表达量则较对照组分别增加了 4 5 %和 33% (P<0 .0 5 ) ,戒断组下降及增加的幅度均低于吗啡组 (P<0 .0 5 ) ;给药 7d后 ,两组 MHC 表达保持在较低水平 ,较对照组分别降低 4 5 .7%和 34.6 % (P<0 .0 5 ) ,而 CD80仍然处于高表达水平 ,较对照组分别增加 5 3%和 4 4 % (P<0 .0 5 )。 结论 :小鼠在吗啡依赖过程中 ,腹腔巨噬细胞 CD80表达显著升高而 MHC 分子表达显著下降 ,可能是导致腹腔巨噬细胞抗原提呈功能下降的原因之?Objective:To explore the changes of CD80 and MHCⅡ expression in peritoneal macrophages of morphine dependent and abstinent mice. Methods:Forty five BALB/c mice were randomly divided into 3 groups:morphine,withdrawl,and control group( n =15).The model of morphine addiction was established by subcutaneous morphine injection in rear limb with increasing doses for 7 d; the withdrawal group was injected with naloxone before morphine injection; the control group was injected with the same volume of saline. Each group was divided into 3 subgroups:3 d injection,5 d injection and 7 d injection group. Then mice were killed and peritoneal cells were harvested by peritoneal lavage with cold PBS buffer for anti CD80 and anti MHCⅡ expression by flow cytometry. Results:On the 3rd day, the MHCⅡ and CD80 expression in morphine group and withdrawl group did not change compared with the control group.On the 5th day, the MHCⅡexpression in morphine group and withdrawl group decreased by 40.4% and 28% compared with that of the control group( P <0.05), and that of the withdrawl group was even lower than morphine group( P <0.05).But the CD80 expression in morphine group increased by 45%( P <0.05) and in withdrawl group by 33%( P <0.05), the latter was lower than the former ( P < 0.05). On the 7th day, the expression of MHCⅡ in morphine and withdrawl group decreased by 45.7% and 34.6% compared with that of the control group( P <0.05). But the CD80 expression of the 2 groups remained increased by 53% and 44% compared with that of the control group( P <0.05).Conclusion: Increased CD80 expression and decreased MHCⅡ expression during morphine addiction may be important factors for depressing macrophage antigen presence function, and it is indicated that the acting is partially via classical opioid receptor.
