摘 要:【目的】观察补阳还五汤对沙土鼠脑缺血及再灌注损伤与脑组织兴奋性氨基酸(EAA)的影响。【方法】复制沙土鼠双侧颈总动脉夹闭脑缺血模型,于缺血后15min再灌注48h,取脑组织测定天冬氨酸(GJu)、谷氨酸(Asp)、γ-氨基丁酸(GABA)、甘氨酸(Gly)的含量。【结果】缺血15min后,脑组织Glu、Asp、GABA含量升高(P<0.05或P<0.01),补阳还五汤可降低脑组织Glu含量(P<0.05);缺血15min再灌注48h后,脑组织Glu及Asp含量升高(P<0.05),补阳还五汤可使脑组织Glu含量降低(P<0.05)。【结论】补阳还五汤抗缺血性脑损伤的作用机理可能与其对抗脑缺血及再灌注后EAA的升高有关。[Objective] To observe the effects of Buyang Huanwu Decoction (BHD) on excitatory amino acid content in brain tissue of gerbils with cerebral ischemia and repeffusion. [Methods] Gerbils were randomized into five groups: A (mimic operation), B (models of ischemia), C (ischemic models treated with BHD), D (models of reperfusion) and E (models of reperfusion treated with BHD). Gerbil models with cerebral ischemia were established by ligation of bilateral common carotid artery. The contents of glurnatate (Glu), asparate (Asp), gama-aminobutyric acid (GABA) and glycine (Gly) in brain tissues were examined after 15 minutes of ischemia and then 48 hours of reperfusion. [Results] After 15 minutes of isehemia, Glu, Asp and GABA contents in brain tissues were increased (P<0.05 or P<0.01 ); BHD could decrease Glu content (P<0.05). After 15 minutes of ischemia and then 48 hours of reperfusion, Glu and Asp contents in brain tissues were increased (P<0.05) and BHD could decrease Glu content (P<0.05). [Conclusion] The mechanism of BHD in counteracting ischemic cerebral injury may be related to the inhibition of increase of excitatory amino acid content in brain tissue.
