摘 要:以培矮 64S和 5个恢复系等 6个两系杂交水稻亲本的DNA为模板 ,进行微卫星分子标记 (SSR)分析 ,结果表明 ,在 60对微卫星引物中有 47对在 6份材料中均有清晰带型扩增产物。以根井标准遗传距离D为亲本间多态率的指标分析 ,培矮 64S与 93 11之间的多态率最高 ,达到 2 6.9% ;其次是与E3 2之间的多态率 ,为 2 5 .9% ;而与特青的多态率最低 (17.0 % ) ,说明培矮 64S与 93 11之间亲缘关系最远 ,而与特青的亲缘关系最近。聚类结果显示 ,微卫星分子标记可将 6份亲本分为 3个大群。认为用SSR标记可以快速有效地鉴定亲本间的亲缘关系 ,从而预测杂种优势 ,为亲本选配提供参考。The microsatellite marker or SSR analysis was conducted, using six two-line hybrid rice parents including (Pei′ai) 64S and five restorer lines as the materials. The results showed that 47 of 60 primer pairs could amplify distinct bands among all parents. Using the Nei′s standard genetic distance D as the index to analyze the polymorphism among parents, it was found that the polymorphic rate between Pei′ai 64S and 9311 was highest at 26.9%, followed by that between Pei′ai 64S and E32 at 25.9% and that between Pei′ai 64S and Teqing was lowest at 17.0%, which indicates that the relationship between Pei′ai 64S and 9311 is farthest while that between Pei′ai 64S and Teqing is nearest. According to the results of the cluster analysis by UPGMA method, the tested six parents were classified into three groups. Group One included Teqing, Shengyou 2 and 219, Group Two contained E32 and 9311, and Group Three had only one parent, i.e., Pei′ai 64S. Therefore, SSR markers can be used to rapidly identify the relationship between parents, so the heterosis of a given F_1 hybrid can be predicted and a good reference can be given to the choice of parents in hybrid rice breeding programs.
