牦牛肿瘤坏死因子-α基因部分片段的克隆测序

摘　　要：根据普通牛和其他哺乳动物基因的碱基序列和相关文献资料设计引物,用PCR扩增并测定了牦牛肿瘤坏死因子-α(TNF-α)基因的部分序列。结果表明,TNF-α基因片段(1160bp)包括外显子3～4、内含子2～3,3'侧翼区和外显子2的部分序列。通过GenBank中的Blastn序列比较分析,牦牛的TNF-α基因与普通牛种同源性高达98%。这为开展牦牛功能基因组计划和分子育种提供了理论依据。In this paper,a partial sequence of TNF-α gene were amplified with PCR and sequenced on \%Bos grunniens.\% The result showed that the TNF-α gene fragment (1160 bp) consisted of exon 3～4, intron 2～3,partial sequence of 3' flanking region and exon 2.The comparison analysis of the fragment sequence with \%Bos taurus \%through blastn of GenBank,the homologies of the nucleotide sequence with \%Bos taurus \%were 98%. This research provide the scientific theoretical foundation on functional gene project and molecular breeding of \%Bos grunniens.
