摘 要:应用SDS电泳及高分辨双向电泳技术,分析了冬小麦经春化、脱春化及超期春化处理后茎类蛋白质组分的变化。发现52.5,38kD和16.2kD蛋白质与冬小麦开花诱导密切相关。mRNA体外翻译结果表明,春化作用中低温诱导与开花紧密相关的mRNA的出现有一定的顺序性。因此推测:春化诱导开花是低温导致某些特定基因表达的结果,而表达的调节主要发生在转录水平上。The molecular events which took place in the shoots of winter wheat during vernalization were investigated in this report. Winter wheat (Triticumaestivurn L. cv Nongda 139) was vernalized at 4℃ for 5 weeks and devernalized at 35℃ for 5 days. Winter wheat without vernalization and spring wheat were use as control, Soluble proteins were extracted and analyzed by SDS-PAGE and two dimensional gel electrophoresis. Total RNA was prepared using phenol/SDS method. Poly(A+) RNA was analyzed by HPLC and translated in vitro in a wheat germ cell-free protein synthesis system. The results showed that low temperature condition altered the soluble protein patterns. Some newly synthesized proteins(52.5, 38 and 16.2 kD) were highly related to the response to vernalization. They were induced by vernalization and disappeared with devernalization and existed in spring wheat without low temperature treatment. There appeared a positive correlation between thesynthesis of these proteins and the ability to flower. Results of in vitro translation showed that mRNA population from vernalized seedlings contained several newly synthesized mRNA species, and some of them remained functional only during a certain period of vernalization. The consistency of the in vitro and in vivo results suggested that changes in gene expression during vernalization occurred most likely at the transcription level.
