摘  要:采用体外渗透和显微注射的方法。将植物微管特效解聚剂甲基氨草磷(APM)引入紫露草雄蕊毛细胞后,发现原来沿着胞质束运动的胞质颗粒运动速度渐慢,进而胞质束消失,颗粒运动停止。显微注射后,还发现APM可通过胞间通道由被注射的细胞向两侧细胞扩散,从而也导致两侧细胞胞质束消失,颗粒运动停止。APM对胞质环流的抑制作用是可逆的。结果表明微管可能是胞质束的重要组份之一,植物胞质环流与微管的聚合与解聚状态有密切关系。Microtubules(MTs) are among the principal components of the cytoskeleton. MTs are also involved in cell movement and transport of materials within cells and in maintaining the shape of cells. Moreover, MTs directly participate in the motion of cilia and flagella ,causing fast axonal transport in animal neurons and provide tracks for rapid movement of particles, including mitochondria.To find out whether cytoplasmic streaming in plant cells is also associated with MTs besides actin and myosin, amiprophos-methyl ( APM ), a specific and effective plant MT inhibitor, has been introduced into the staminal hair cells by immersion and microinjection. Permeation of APM into the cells caused the slowing down of particle movement along cytoplasmic strands, the dissociation of the particles from the strands, and, finally, the stopping of the particle movement. After being microinjected, APM could spread to the cells adjacent to the injected cell through intercellular channels, causing the disappearance of the cytoplasmic strands and the stopping of the particle movement. In 30 min, however, the cytoplasmic strands reappeared in the cells on the far end from the injected cell, then the particle movement started again. Therefore the inhibition by APM is found to be reversible. The above-mentioned results indicate that MT may be a principal component of cytoplasmic strands and that there is a close relationship between the cytoplasmic streaming and the polymerization and depolymerization of the MTs.
