摘  要:目的 探讨葛根素对模拟急性低气压环境下噪声性听器损伤的防治作用。方法 将 6 0只听力正常豚鼠随机分为正常组、阳性对照组、葛根素防治组和葛根素治疗组。后 3组置于模拟高原海拔 5 5 0 0m低气压环境 ,130dBSPL白噪声持续刺激 8小时 ;防治组和治疗组每日腹腔注射葛根素注射液 (15 0mg/kg)。观察各组的听性脑干反应 (ABR)反应阈、耳蜗形态学改变、血液流变学和血清丙二醛 (MDA)、超氧化物岐化酶 (SOD)含量的变化。结果 出舱后 8小时 ,各组ABR阈移最大 ,对照组、防治组在 1kHz的阈移分别为 33.6 7± 5 .4 0、2 6 .5 0± 4 .76dB ,在 8kHz的阈移分别为 5 9.33± 5 .2 0、4 8.6 7± 4 .70dB(P <0 .0 1) ;高频听力损失较低频严重 (P <0 .0 1)。以后各组ABR阈移逐渐恢复 ,给药两组恢复程度均较对照组明显 (P <0 .0 1)。形态学改变与听力变化一致。防治组血清MDA含量显著低于对照组 ,用药的两组血清SOD均高于对照组。对照组血液流变学全血粘度值明显升高 ,与用药两组比较有显著性差异。结论 葛根素对急性低气压环境下的听器损伤具有一定的防治效果 ,可能与其改善微循环和血液流变学特性。Objective To investigate the effects of purarine on noise deafness in acute hypobaric and hopoxic environment.Methods Sixty guinea pigs were randomly distributed into 4 groups (normal group, control group, Pur-prophylactico-therapeutic group, and Pur-treated group). The last three groups were exposed to 130 dB white noise in modeling low pressure environment of 5 500 m which successively lasting for 8 hours. The last two groups were treated by intraperitoneal injection with purarine at the dosage of 150 mg·kg -1 per day. Acoustic brainstem response(ABR), the surface preparation and transmission electron microscope were utilized to evaluate the hearing thresholds and cochlear morphology. To explore the mechanism associated with Purarine protection, hemorheology index together with serum levels of MDA and SOD were measured.Results Mean threshold shifts at 1 kHz & 8 KHz of control group on the 8 th hour after exposure developed up to 33.67±5.40 and 59.33±5.20 dB SPL, while the mean threshold shifts of Pur-intervening group developed only 26.50±4.76 and 48.67±4.70 dB SPL( P <0.01). The threshold shifts at high frequencies were significantly larger than that at low( P <0.01).Then the hearing of all groups recovered gradually, and the two Pur-utilized groups recovered more rapidly than control group ( P <0.01). Morphological changes were consistent with functional changes. MDA concentration in serum of Pur-intervening group was significantly lower than that in control group( P <0.01). SOD levels of the two Pur-utilized groups were much higher than that in control group ( P <0.05). The hemorheological index of whole blood viscosity of control group increased notablely comparing with the Pur-utilized groups.Conclusion This study suggests that Purarine has certain protective effect on hearing loss induced by noise in modeling acute hypobaric and hypoxic environment.
