摘 要:目的 :探讨芥子气中毒早期肝组织内皮素_1(ET_1)、内皮素A型受体 (ETAR)的变化和意义。方法 :应用免疫组化及RT_PCR技术 ,观测大鼠芥子气中毒后肝组织ET的含量及ET_1,ETARmRNA水平的变化 ,以及该变化与肝组织损伤的关系。结果 :中毒后肝组织ET的含量先下降后增加 ,12h达峰值 ;中毒后肝preproET_1mRNA的表达在 2~ 6h低于对照组 ,12h增加达峰值 ;ETARmRNA的表达没有明显的变化。结论 :ET_1及其受体ETAR在芥子气中毒后肝的病理生理过程中可能起着重要作用。Objective:To explore the role of endothelin-1(ET-1) and endothelin A receptor(ETAR) in pathophysiology of the liver after mustard poisoning.Methods:The changes in transcription and expression of ET-1 and ETAR in the liver and their relationship with the tissue damage were studied by immunohistochemistry and RT-PCR. Results: The level of ET in the liver was significantly decreased 2-12 hours after mustard poisoning, but increased at 12 hours. The transcription and expression of ET-1 was decreased at the early stage after poisoning, but increased at 12 hours. mRNA of endothelin A receptor was unchanged significantly. Conclusion: ET-1 and ETAR may play important roles in the pathophysiological process of the liver after mustard poisoning.
