复方丹参滴丸对2-乙酰氨基芴致突变性的影响

摘　　要：目的　观察复方丹参滴丸诱导处理对原致癌物 2 乙酰氨基芴 (2 AAF)致突变作用的影响。方法　将复方丹参滴丸不同剂量诱导处理后的大鼠肝S9作为代谢活化系统进行Ames试验 ,比较加入S9前后 2 AAF对鼠伤寒沙门氏菌的回复突变率。雄性昆明小鼠以不同剂量复方丹参滴丸诱导处理 ,比较未诱导 (- )与诱导 (+)处理后 ,2 AAF引起骨髓微核率及PCE/NCE比值的变化。结果　经复方丹参滴丸不同剂量诱导处理的大鼠肝S9复 和空白对照肝S9对 对 2 AAF均有代谢活化作用 ,TA98、TA10 0菌株的菌落回变数比无S9活化时增高 5倍左右 ,但两两比较无明显差异。微核试验表明 2 AAF引起小鼠骨髓微核率明显升高 ,PCE/NCE比值下降。复方丹参滴丸三个不同剂量诱导处理后 ,2 AAF致小鼠骨髓微核率与未用复方丹参滴丸诱导处理时相比两项指标均无明显差异 ,复方丹参滴丸三个剂量诱导组间也无明显差异。结论　复方丹参滴丸未见致突变作用 ,用其诱导处理 ,亦不能增强 2Aim To observe the effect of Compound Danshen Dripping Pills on the mutagenicity of 2-acetamide-fluorene(2-AAF).Methods Ames test was conducted with rat liver S9 metabolic activation system(S9-mix) induced by Compound Danshen Dripping Pills at different doses,and the amounts of reverse mutation induced by 2-AAF were compared between groups with different S9-mix.Male Km mice were induced with Compound Danshen Dripping Pills at different doses followed by 2-AAF ip.The frequencies of micronucleus in polychromatic erythrocytes in bone marrow were compared between groups with and without Compound Danshen Dripping Pills induction.Results 2-AAF could be activated by rat liver S9-mix induced by Compound Danshen Dripping Pills at three different doses and negative control.The amounts of reverse mutation on strain TA98 and TA100 induced by 2-AAF with four S9-mix were five times more than those without S9-mix.But there is no difference between any two groups above.Micronucleus test showed that the micronucleus frequencies in mice bone marrow induced by 2-AAF increased significantly while PCE/NCE ratios declined significantly.The micronucleus frequencies in mice bone marrow and PCE/NCE ratios induced by 2-AAF showed no statistical difference between groups with and without Compound Danshen Dripping Pills induction at different doses,and both indexes didn't change significantly among Compound Danshen Dripping Pills induced groups at three doses.Conclusions The mutagenicity of 2-AAF couldn't be enhanced by Compound Danshen Dripping Pills induction.
