摘 要:目的 :从细胞因子基因调控研究二磷酸盐防治人工关节松动的作用机理。方法 :分离培养人外周血单个核细胞 ,加入关节磨屑及不同浓度的阿伦膦酸钠 ,分别用原位杂交及逆转录聚合酶链式反应 (Reversetranscription polymerasechainrectionRT PCR)技术检测IL 6mRNA的表达。结果 :关节磨屑组IL 6mRNA的表达显著高于对照组 (P <0 .0 1) ,不同浓度的阿伦膦酸钠组IL 6mRNA的表达明显低于关节磨屑组 (P <0 .0 1)。结论 :关节磨屑刺激人外周血单个核细胞IL 6mRNA的表达 ,阿伦膦酸钠能通过下调人外周血单个核细胞IL 6mRNA的表达 ,对人工关节松动可能起防治作用。Objective:To study the mechanism of bisphosphonates in the prevention and treatment of aseptic loosening of prosthesis on cytokines-related genes modulation.Methods:In vitro human peripheral blood mononuclear cells(PBMC) were cultured with wear particles and sodium allendronate of different concentration.Then expression of IL-6m RNA was determined by In Situ Hybridization and RT-PCR techniques.Results:The expression of IL-6mRNA was increased significantly when wear particles was added as compared with control group ( P <0.01).Hybridization and RT-PCR demonstrated that bisphosphonates reduced IL-6mRNA expression.Conclusion:Wear debris can stimulate IL-6mRNA expression of human PBMC.Bisphosphonates may be used in the prevention and treatment of aseptic loosening of prosthesis by downregulating IL-6mRNA expression of human PBMC.
