摘  要:目的探讨家兔坐骨神经细胞外基质 (extracellularmatrix ,ECM )的制备 ,并观测其生物相容性 ,为临床应用ECM修复神经干缺损提供实验依据。方法采用本室改良的NaOH消蚀技术 ,制备去除细胞成分的家兔坐骨神经ECM支架 ,进行光镜和扫描电镜观察和分析。将制备的ECM包埋于同种异体家兔背部肌肉组织中 ,术后不同时间取出 ,行大体及光镜下组织学观察 ,检测其有无排异反应及周围血管长入情况。结果①经NaOH消蚀法处理的坐骨神经组织细胞成分被完全消蚀掉 ,神经外膜、神经束膜和神经内膜仍保持原有的构筑特征。②肌肉包埋后 ,宿主的成纤维细胞和血管内皮细胞向ECM内生长 ,形成较多富含红细胞的血管 ,并可引导再生细胞定向排列。结论本室改良的NaOH消蚀法制备的周围神经ECM神经膜管结构保存完好 ,有良好的组织相容性 。ObjectiveTo explore the preparation method of the nerve extracellular matrix(ECM),evaluate its biocompatibility in rabbits and thereby supply an experiment foundation to clinical application. MethodsThe materials were made by immersing the samples of sciatic nerve into NaOH aqueous solution and were cleaned by ultrasonic wave(USW),then observed and analyzed by light microscope and scanning electron microscope(SEM).The material was implanted into the muscles of rabbits for the study of biocompatibility. Results① The cellular elements of the sciatic nerve treated with NaOH maceration were eliminated completely; the endoneurium, perineurium and epineurium were complete and remained three dimensional structure. ② The fibroblasts and endotheliocytes grew into the ECM, and formed blood vessels. ConclusionThe ECM made with this method can reserve original three dimensional structure. It has a good biocompatibility and can induce arrangement of the host regenerative cells directionally.
