摘 要:通过检测去卵巢大鼠给予补肾中药后腰椎内ERmRNA及OPGmRNA的表达 ,进一步探讨补肾中药防治骨质疏松的分子机制。将雌性 3月龄SD大鼠 4 8只 ,随机分为卵巢切除 +补肾中药防治 (HDP) ,卵巢切除 +雌激素 (倍美力 )防治 (ER) ,骨质疏松(卵巢切除 ) (OVX)及正常对照组 (SHAM) 4组 ,每组 12只。术后 12周处死大鼠 ,取L3 椎体 ,异硫氰酸胍一步法提取总RNA ,嵌套式反转录聚合酶链反应 (NRT PCR) ,扩增待检测各组ERmRNA及OPGmRNA的表达。同时取L4椎体 ,行骨组织形态计量学检测。结果显示HDP组ERmRNA及OPGmRNA阳性表达率分别为 75 .0 %和 6 6 .7% ;EP组ERmRNA及OPGmRNA阳性表达率均为 83.3% ;OVX组各例ERmRNA均无表达 ,2例OPGmRNA呈弱阳性表达 (阳性率为 16 .7% ) ;SHAM组呈ERmRNA及OPGmRNA阳性表达。χ2 检验 ,HDP组与ER组比较ERmRNA及OPGmRNA阳性表达率均无显著性差异 (P >0 .0 5 ) ;该两组与OVX组比较 ,ERmRNA及OPGmRNA阳性表达率均有高度显著性差异 (P <0 .0 1) ;而与SHAM组比较 ,HDP组ERmRNA ,ER组ERmRNA及OPGmRNA阳性表达率无显著性差异 (P >0 .0 5 ) ,但HDP组OPGmRNA阳性表达率有显著性差异 (0 .0 1<P <0 .0 5 )。骨组织形态计量学检测 ,OVX组骨体积、骨小梁厚度及骨小梁数目均比SHAM组显著减少 ,而骨小梁?The objective of the paper is to further explore the molecular mechanism of kidneys-tonifying Chinese meteria medica (KTCMM) to prevent and treat osteoporosis through examining estrogen-receptor- (ER-) and osteoprotegerin- (OPG-) mRNA expressions in the lumbar vertebrae of ovariectomized rats given KTCMM. Forty-eight female SD rats aged three months were randomly divided into four groups: Group I ovariectomized and given KTCMM; Group II ovariectomized and given estrogen-Premarin; Group III osteoporosis (ovariectomized and given saline); and Group IV normal control (12 rats each group). The rats were killed 12 weeks postoperatively, the L3 specimens were taken, the total RNAs were extracted by one-step method of guanidinium isothiocyanate, the ER- and OPG- mRNA expressions of each group were examined by means of NRT-PCR (nested reverse transcriptase polymerase chain reaction). At the same time, the L4 specimens were taken and analyzed by histomorphometry. The results showed: (1) the positive rates of ER- and OPG-mRNA expressions of Group I were 75.0 % and 66.7 %, respectively; (2) the positive rates of both ER- and OPG-mRNA expressions of Group II were 83.3 %; (3) Group III had no ER- mRNA expression and only weak positive OPG-mRNA expression with a positve rate of 16.7 % in two cases; (4) Group IV had positive expressions of both ER- and OPG-mRNAs. χ2 test showed: (1) no significant difference of Group I from Group II in the positive rates of ER- and OPG-mRNA expressions (P > 0.05); (2) a very significant difference of Group I and Group II from Group III in the positive rates of ER- and OPG-mRNA expressions (P < 0.01); (3) no significant difference of Group IV from Group I in the positive rate of ER-mRNA expression and from Group II in the positive rates of ER- and OPG-mRNA expressions (P > 0.05) but a significant difference of Group IV from Group I in the positive rate of OPG-mRNA expression (P < 0.01 or P < 0.05).
