醋酸精氨酸的毒理学试验研究

摘　　要：目的　通过急性毒性实验和长期毒性实验 ,观察醋酸精氨酸对动物的毒性反应。方法　急性实验选用KM小鼠 80只 ,随机分成 4组 ,每组 2 0只 ,两个组分别从尾静脉注射 0 5g/ml醋酸精氨酸和盐酸精氨酸 2 5ml/kg体重 ;另两个组分别一次灌胃 0 8g/ml的醋酸精氨酸和盐酸精氨酸 4 0ml/kg体重 ,观察 7d。长期毒性实验选用Wistar大鼠 6 0只 ,分高、低剂量及正常对照组3个组。杂种狗 6只 ,分高剂量组和正常对照组 ,高剂量组大鼠以 2 0 0 g·kg-1 ·d-1 、狗 1 2 0 g·kg-1 ·d-1 ,低剂量大鼠组 2 0 g·kg-1·d-1 ,观察 4周。结果　小鼠急性试验 (静脉和灌胃 )所用剂量远大于拟用临床剂量的 (0 4g·kg-1 ·d) 80倍以上 ;长期毒性试验采用小鼠 5 0倍于临床拟用剂量 (2 0 g/kg/d) ,狗 30倍于临床拟用剂量 (1 2 0 g/kg/d) ,未发现明显的中毒反应 ,也未发现主要靶器官的损害。结论　通过急性和长期毒性实验 。Objective To investigate toxicity response and damage of main target organs of acetic arginine,in the immediate and long term toxicity experimernt in animal.Methods Eighty KM mouse were divided into 4 groups in immediate toxicity experiment .Two groups were injected with acetic arginine or hydrochloric arginine in 12.5g/kg body weight through tail rein respectively;and the other two groups were pouring feeding with acetic arginine or hydrocholric arginine in 32.0g/kg body weight respectively observed , lasting 7 days.In the long term toxicity experiment:60 rats were divided into 3 groups:one takes acetic arginine 2.0g/k/d,the other takes 20.0g/k/d,another group as control respectively;6 dogs were divided into 2 groups on takes 12.0g/k/d,the other is control.The experiment lasted 4 weeks.Results The result illustrated:quantity used in mouse by vein injection and pouring feeding as immediate toxicity experiment was far larger than the intending clinical dosage(0.4g/kg body weight),it was 80 times above;in long term toxicity experiment the amount (12.0g/kg/d)it was taken more than 30 times of its intending clinical dosage in dogs,and more than 50 times(20.0g/kg/d)in mouse,toxicity response and damage of main target organs were not appeared.Conclusion Acetic arginine is safe in the immediate and long term toxicity experiment in animal.
