雌激素对去卵巢大鼠血清瘦素的影响

摘　　要：目的:探讨雌激素对去卵巢大鼠血清瘦素水平的影响。方法:40只健康雌性未交配6个月龄SD大鼠用去卵巢手术制作动物模型,分为4组:假手术组、雌激素一组、雌激素二组、手术组;去卵巢手术后1周开始雌激素一组皮下注射雌激素20μg/(kg·d),共12周,雌激素二组在处死前2d开始皮下注射雌激素40μg/(kg·d)。动物处死前测量体质量,并留取血清标本,进行血清瘦素水平分析。结果:血清瘦素水平(μg/L)手术组(8.82±0.98)和雌激素二组(17.03±2.70)较假手术组(14.50±1.71)和雌激素一组(9.66±1.27)明显增高(t=9.014,8.11,P均<0.01),雌激素二组较手术组明显升高(t=2.494,P<0.05),假手术组和雌激素一组之间无明显差异(t=8.11,P>0.05)。体质量(g)手术组(317±22)和雌激素二组(369±77),假手术组(374±33)和雌激素一组(309±31)差异无显著性意义(t=0.795,0.439,P均>0.05)结论:雌激素可以促进去卵巢大鼠瘦素的分泌,卵巢切除术后大鼠血清瘦素水平较假手术组明显升高,可能是体质量增加的结果。AIM:To explore the effect of estrogen on level of serum leptin in ovariectomized rats. METHODS:Forty healthy virgin female SD rats of 6 months old were divided into 4 groups:sham group;estrogen group 1[17β estradial was injected at the dosage of 20 μg/(kg·d) 1 week after operation and continued for 12 weeks; estrogen group 2[17β estradial was injected at the dosage of 40 μg/(kg·d) only 2 days before sacrifice]and operation group.Body mass was measured before sacrifice,and serum samples were collected to determine the leptin level. RESULTS:Serum leptin concentration(μg/L) in the operation group(8.82±0.98) and estrogen group 2(17.03±2.70) were increased more significantly than those in the sham group(14.50±1.71) and estrogen group 1(9.66±1.27)(t=9.014, 8.11,P< 0.01),it was obviously higher in estrogen group 2 than in the operation group(t=2.494,P< 0.05),and no significant difference between the sham group and estrogen group 1(t=8.11,P >0.05).There was no significant difference in body mass(g) among operation group(317±22),estrogen group 2(369±77),estrogen group 1(309±31) and sham group(374±33)(t=0.795,0.439,P >0.05). CONCLUSION:Estrogen can promote the secretion of serum leptin of ovariectomized rats.Serum leptin level increases obviously in the sham group,which may be the result of increase of body mass.
