摘 要:【目的】探讨系统性红斑狼疮(SLE)患者骨髓基质细胞上清液对造血干(祖)细胞的作用及其机制。【方法】半固体集落培养法观察28例活动期SLE骨髓基质细胞生长及其上清液对正常细胞、自身骨髓干/祖细胞的抑制作用;检测SLE其骨髓基质细胞上清液中TNF-α、IFN-γ的水平;RT-PCR法检测骨髓基质细胞IFN-γmR-NA的表达。【结果】①29%活动期SLE患者骨髓基质细胞的集落数较正常人明显减少(P<0.05)。②体积分数为15%的活动期SLE骨髓基质上清液对正常人骨髓、自身SLE造血干/祖细胞混合集落(CFU-Mix)的形成较对照组具有明显抑制作用(P<0.05)。③活动期SLE患者骨髓基质细胞的上清液TNF-α、IFN-γ的水平高于稳定期SLE组、正常对照组(P<0.05)。④70%活动期SLE患者骨髓基质细胞有IFN-γmRNA的表达,而正常对照则未见有IFN-γmRNA的表达。【结论】活动期SLE患者骨髓基质细胞上清液对正常、自身骨髓造血干(祖)细胞的有抑制作用,这与其基质细胞存在高表达TNF-α、IFN-γ有关。To investigate the suppressive effect of cultured supernants of bone marrow stromal cells(BMSC) from active systemic lupus erythematosus(SLE) patients on haematopoietic progenitor cells and its mechanism. The colony formation of stromal cells was examined in 28 patients with active SLE in vitro using methycellulose culture .The suppression of mix colony formation units (CFU Mix) of BMSC from healthy individuals and SLE was studied in vitro with cultured supernatants of BMSC from 28 active SLE patients .Cytokine IFN γand IFN αwere measured in culture supernatants with SLE by using enzyme linked immunosorbent assay(ELISA).The IFN γmRNA expression of marrow stromal cells from patients with systemic lupus erythmatosus was assayed by a semiquantitative reverse transcriptase polymerase chain (RT PCR). ①The number of colonies formation stromal cells from 29%patients with active SLE was lower than that healthy individual(P< 0.05). ②Cultured supernatants of BMSC from 28 active SLE patients at contained 15%was significantly more suppressive to allogeneic and autologous CFU Mix growth than that of BMSC from control (P< 0.05). ③The TNF αor INF γlevel in cultured supernatants of BMSC from 28 active SLE patients was higher than that from inactive SLE and healthy control(P< 0.05). ④The expression of mRNA IFN γtranscripts was significantly increased in BMSC of 70%patients with active SLE compared with healthy controls (P< 0.01).[Conclusion]Cultured supernants of bone marrow stromal cells from active SLE patients may suppress the formation of allogeneic and autologous CFU Mix, which were related to high level IFN γand IFN αin bone marrow stromal cells of SLE patients
