摘　　要：白细胞介素 2 4 (IL- 2 4 )又称黑素瘤分化相关基因 7(melanom a differentiation associated gene- 7,MDA- 7)主要表达于脾、胸腺、外周血白细胞、单核细胞等免疫组织和分化的黑素细胞 ,其他组织细胞可被诱导表达。 IL- 2 4受体有 2种 (IL- 2 2 R1 /IL- 2 0 R2和 IL- 2 0 R1 / IL- 2 0 R2 )。 IL- 2 4可诱导人外周血单个核细胞产生高水平 IL- 6、TNF- α和 IFN- γ,以及少量 IL- 1 β、IL- 1 2和粒 -巨噬细胞集落刺激因子 (GM- CSF)。将携 IL- 2 4基因的腺病毒质粒 (Ad- IL- 2 4 )转染细胞后能抑制多种肿瘤细胞的增殖并诱导细胞凋亡 ,但对正常细胞无影响。IL- 2 4的这些作用与其诱导凋亡相关因子 (BAX/ Bcl- 2 ,BAK/ Bcl- 2 )的比例变化、上调p38- MAPK及可被生长停滞和 DNA损伤诱导的基因家族产物增加有关。提示 IL- 2 4有治疗肿瘤的临床价值 ,基于 Ad- IL- 2 4基因治疗的临床试验已经开始。现就 IL- 2Interleukin 24(IL 24),also called the melanoma differentiation associated gene 7(MDA 7),is stably expressed in human tissues associated with the immune system such as the spleen,thymus,peripheral blood leukocytes and normal melanocytes. IL 24 binds to IL 20 and IL 22 receptor complexes(IL 22R1/IL 20R2 and IL 20R1/IL 20R2),and induces secretion of high levels of IL 6,TNF alpha,and IFN gamma and low levels of IL 1beta,IL 12,and GM CSF from human PBMC. Adenoviral IL 24(Ad IL 24) induces growth suppression and apoptosis selectively in diverse human cancers and tumor cell lines without producing any apparent harmful effect in normal cells. The effects of Ad IL 24 are associated to the ratio of pro apoptotic(BAX,BAK) to anti apoptotic(Bcl 2) proteins,and the up regulation of p38 MAPK and a family of growth arrest and DNA damage(GADD) inducible genes. These demonstrate the potential therapeutic effects of Ad IL 24 in human cancer.
