摘  要:用新城疫病毒(NDV)F48E8株作为包被抗原,建立了检测由鸡痘病毒载体介导表达的NDV融合蛋白(F)抗体的间接ELISA方法。通过对20份阳性血清的P/N值与ELISA抗体效价(ET)的自然对数值(lnET)的线性回归分析,获得了回归方程lnET=0.201×P/N+4.632。应用该ELISA方法对表达NDVF基因的重组鸡痘病毒免疫鸡群进行了血清抗体检测,结果表明,血清的ELISA抗体效价与相应血清中和试验的抗体效价高度相关。攻毒保护试验的结果显示,ELISA抗体效价与机体的保护力表现一定程度的相关性。An indirect ELISA for detecting antibody to the fusion protein of Newcastle disease virus (NDV) expressed in recombinant fowlpox virus was developed with great sensitivity, high specificity and strong reproducibility. A near-linear relationship between positive ratio and negative ratio of anti-sera at a single working dilution (1∶40). Regression analysis was used to construct a standard curve. An equation lnET =0.201×P/N+4.632 was obtained from the regression analysis, which allow to rapidly predict ET. In a comparative (study), a strong correlation (P<0.01) was found between ELISA titer (ET) and (virus-)neutralization antibody titer of vaccinated chickens against fusion protein of NDV. Subsequently, (experiment) on chickens challenged with virulent NDV strain showed that protection seems to be correlated with the ELISA antibody (titer).
