摘 要:目的 探讨 hhlim蛋白在心肌肥厚发生中的地位和作用。方法 建立腹主动脉部分结扎心肌肥厚模型和运动性心肌肥厚模型 ,应用RT- PCR和 Western blot等方法观察 hhlim在生理性和病理性心肌肥厚发生中的作用。结果 运动组和腹主动脉结扎组动物的心脏重量、心系数、收缩压、舒张压和平均动脉压较对照组明显增大 ,说明模型建立是成功的。RT- PCR和 Western blot结果显示 ,对照组大鼠心肌细胞 hhlim表达较低 ,运动组和腹主动脉结扎组大鼠 hhlim表达明显增加 ,以腹主动脉结扎组增加最明显。结论 无论是运动性还是高血压性心肌肥厚 ,虽然其引发机制及信号转导通路不同 ,但都与激活 hhlim的表达有关。Objective To investigate the function of hhlim in cardiac hypertrophy. Methods We established the pressure-overloaded hypertension and swimming- overloaded cardiac hypertrophy models in rats. The expression of hhlim was assayed by RT-PCR and Western blot. Results The LV weight/body weight and blood pressure of experimental group increased obviously compared with those of control group, which suggested that abdominal aorta binding and swimming had made LV pressure-overloaded cardiac hypertrophy. Compared with low-level expression of hhlim in cardiac muscle of controls, the contents of hhlim mRNA and protein increased significantly in experimental group. The expression of hhlim in abdominal aorta binding group increased dramatically than that of the swimming group. Conclusions Although the mechanism of swimming-overloaded cardiac hypertrophy may be different from that of hypertensive hypertrophy, the activation of hhim is participated in the course of cardiac hypertrophy induced by the abdominal aorta binding and swimming.
