摘  要:目的 探讨雌激素对去卵巢大鼠记忆功能、海马部位 β淀粉样蛋白 1 - 4 0 (Aβ1 - 4 0 )及蛋白激酶 C(PKC)表达的影响。方法 大鼠去卵巢建立动物模型 ,1 7w行水迷宫试验 ,1 8w取脑组织做 Aβ1 - 4 0及 PKC的免疫组化染色。结果 去卵巢大鼠的雌激素 (E2 )水平明显降低 (P<0 .0 1 ) ;去卵巢组的记忆功能下降 ;脑海马部位的 Aβ1 - 4 0表达比假手术组和雌激素替代组增加 (P<0 .0 1 ) ,而 PKC表达则减少 (P<0 .0 1 )。结论 雌激素可能是通过提高 PKC的表达水平 ,使淀粉样前体的代谢生成 Aβ1 - 4 0减少来保护海马部位胆碱能神经元 ,从而改善记忆功能。雌激素可能是影响记忆功能重要因素之一。Objective To investigate the effects of estradiol on memory function and the expression of amyloid-beta protein 1-40 (Aβ1-40) and protein kinase C (PKC) in the hippocampus of ovariectomized mice. Methods Using the animal model of ovariectomized mice, we detected the memory function of rats by morris water maze in the 17th week and got the brain to do immunohistochemical staining of Aβ1-40 and PKC in the 18 th week. Results The level of estradiol was lower (p<0.01) and the memory function was weaker in the ovariectomized group significantly. Aβ1-40 immunoreactivity positive neurons distributed abundantly but PKC immunoreactivity positive neurons reduced in hippocampus in the ovariectomized mice compared with the other two groups. Conclusions Estradiol can improve the memory function maybe by increasing the expression of PKC to reduce the production of Aβ1-40 protein. Estradiol may be one of the important factors for memory function.
