摘　　要：目的 :　克隆人壳三糖酶 (chitotriosidase)基因并进行序列分析。方法 :　分离和培养人外周血单核巨噬细胞 ,提取总RNA ,反转录成cDNA ,通过PCR方法扩增壳三糖酶编码基因 ,克隆至pUC19载体 ,测序并分析其序列。结果 :　所克隆的壳三糖酶基因在外显子 10处没有 2 4个碱基的重复序列。结论 :　该壳三糖酶基因属于野生型 ,在抗真菌病方面 ,可能有着重要而广阔的临床应用前景。Objective: To clone and analyze Homo sapiens chitotriosidase gene. Methods:Total RNA was isolated from cultured human macrophages and mRNA was reversely transcribed into cDNA. Then PCR was used to amplify the chitotriosidase gene. The PCR product was cloned into pUC19 plasmid and sequenced. Results: There was no 24-base pair duplication in exon 10 in our cloned gene. Conclusion :The coloned chitotriosidase gene is a wild type.This may signify an important and significant prospect in antifungal application.
