施万细胞和MK801对大鼠脊髓半横切后背核神经元存活及其表达NOS的影响  被引量:9

摘  要:目的 观察单独或联合应用施万细胞和MK80 1对大鼠脊髓半横切后受损伤的背核神经元存活及其表达NOS的影响。 方法 应用细胞培养、免疫组织化学、核荧光标记、荧光逆行追踪和酶组织化学等技术。 结果 T11脊髓半横切后 15d ,生理盐水对照组的L1脊髓段损伤侧背核神经元的存活数量比未损伤侧减少 ,其中胞体面积 >4 0 0 μm2 的神经元明显减少 ,而且部分存活的神经元NOS活性增高 ,背核面积及神经元胞体面积缩小。施万细胞组、MK80 1组、施万细胞与MK80 1联合组的L1脊髓段损伤侧背核神经元的存活数量则比术后同期对照大鼠增多 ,胞体面积 >4 0 0 μm2 神经元的存活数量显著增多。施万细胞与MK80 1联合组作用明显。MK80 1组存活的神经元中NOS活性明显减弱 ,且能阻止背核及其神经元胞体的面积的缩小。 结论 施万细胞与MK80 1均可促进轴突被切断的背核神经元的存活 ,两者有协同作用 ,且MK80Objective To observe the effects of Schwann cells(SCs)and MK801 used respectively or jointly on the neuronal survival and expression of nitric oxide synthase(NOS)in Clarke's nucleus(CN)after spinal cord hemisection(SCH) of rats. Methods Cell culture,immunohistochemistry,nucleus fluorescence labeling,fluorescence retrograde labeling and enzymatic histochemistry technique were used. Results The number of survived CN neurons on the lesioned side at L1 spinal segment,compared with that on the intact side,decreased significantly 15 days after T11 spinal cord hemisection of vehicle group,particularly those whose soma size is more than 400μm 2.The activity of NOS could be detected in some survived neurons.Besides,the mean soma area of CN neurons on the lesioned side was significantly smaller than that on the intact side.The number of survived CN neurons on the lesioned side in L1 spinal segment of SCs group,MK801 group,SCs and MK801 joint group 15 days after T11 spinal cord hemisection was increased than that of vehicle group in the same period, particularly those whose soma size is more than 400μm 2.The effect of MK801 and SCs joint group was significant.The NOS activity in survived CN neurons was decreased and the shrinkage of their soma size was prevented in MK801 group.Conclusion\ SCs and MK801 can promote the survival of CN neurons which are axonotmesis.There is a cooperative effect between SCs and MK801.MK801 may also prevent the shrinkage of CN and its neuronal soma.\;[
