摘  要:目的  制备神经生长因子 (NGF)抗体 ,并初步探讨其应用价值。 方法  将NGF免疫新西兰兔 ,经 5次加强免疫 ,得到了高效价的NGF抗血清 ,并建立了相应的放射免疫分析 (RIA)方法。结果 抗血清滴度为 1∶2 0 0 0 0。亲和力K =1.6 6× 10 10 mol/L。抗血清特异性强 ,与GDNF ,CNTF ,EGF ,β EP ,CCK ,GHRH ,GH ,BNP ,AVP ,Bombesin等均无交叉反应。抑郁症患者血浆NGF浓度为 (36 0 .0 5± 5 1.89)pg/ml,正常人为 (313.5 3± 34.0 2 )pg/ml(P <0 .0 0 1)。小鼠颌下腺 (每mg组织湿重 )NGF含量为 (2 0 2 .0 9± 32 .6 1)ng。 结论  该抗血清滴度高、亲和力强、特异性强 ,建立的NGF放射免疫分析方法灵敏度高、操作简便 ,为进一步研究NGF在生理和病理状态下的作用提供了可靠的方法。Objective To prepare nerve growth factor (NGF) antiserum and investigate its primary application. Methods NGF was used to immunize the New Zealand rabbit and antibody against NGF was prepared. The high titre of antiserum was gained and radioimmunoassay (RIA) for NGF detecting radioimmunoassay (RIA) was established. Results The NGF antiserum possesses high titre (1∶20000) and high affinity (K=1.66×10 -10 mol//L ) . The specificity of NGF antiserum was high and it did not react with GDNF, CNTF, EGF, β-EP, CCK, GHRH, GH, BNP or Bambsin. The plasma level of NGF in depression people was (360.05±51.89) pg/ml while that of control was (313.53±34.02) pg/ml . The NGF level in mouse submaxillary glands was (202.09±32.61) ng/mg. Conclusion The NGF anti-serum we gained possesses high titre, high affinity and high specificity. RIA of NGF we established is delicate, simple and rapid. It is a reliable methed for studying the role of NGF in various physiological and pathophysiological conditions.
