刺五加对人胚肺二倍体纤维母细胞生长的影响

摘　　要：目的 :探讨刺五加 (APS)对人胚肺二倍体纤维母细胞 (2 BS)增殖、凋亡的影响。方法 :采用生物活性法观察不同浓度APS在不同时间对 2 BS细胞增殖的抑制作用 ,利用流式细胞仪检测其诱导 2 BS细胞凋亡小体形成百分率及对细胞周期的影响 ,并与甲基强的松龙 (M- pred)比较。结果 :1APS和 M- pred对 2 BS细胞均有增殖抑制作用 ,与无药空白对照组比较差异有统计学意义 (P <0 .0 1) ,但两药作用相比差异无统计学意义 (P >0 .0 5 ) ;2 APS和 M- pred均有诱导 2 BS细胞凋亡作用 ,主要作用于 S期 ,2 BS细胞凋亡小体形成百分率与无药空白对照组比较差异均有统计学意义 (P <0 .0 1) ,但两药作用相比差异无统计学意义 (P >0 .0 5 )。结论 :APS有显著抑制 2 BS细胞增殖并诱导其凋亡的作用、活性与 M- pred相近 ,但由于 APS尚有多种药理作用而无不良副反应 ,所以是治疗肺纤维化较好的药物。Objective:To investigate the effects of APS on the proliferation and apoptosis of 2BS cells.Methods:Compared with the Methylprednisolone (M-pred),the effects of different concentration APS in different duration on the proliferation of 2BS cells were evaluated by bioassay and the apoptosis body ratio induced by the APS were analyzed by flow cytometry.Result:①M-pred and APS have similar strong proliferative inhibition on the 2BS cells (P>0.05), and have significant difference comparing with untreated control group. ②M-pred and APS markedly induced apoptosis in 2BS cells (P<0.01) and acted chiefly in S period, but no different effect was found between the 2 drug (P>0.05).Conclusion:M-pred and APS significantly inhibit the proliferation of 2BS cells and promote the apoptosis of these cells ,and because of its multi-effect and has no side effect, APS may be an ideal option in the treatment of pulmonary fibrosis.
