健胃愈疡颗粒剂对大鼠胃粘膜的保护及其作用机制探讨

摘　　要：目的探讨健胃愈疡颗粒 (JWYY)对胃溃疡大鼠粘膜的保护作用及其可能的机制。方法用Okabe改良法复制SD大鼠胃溃疡模型 ,用半定量逆转录多聚酶链反应 (RT—PCR)检测转化生长因子 —β1(TGF—β1)和白细胞介素 —8(IL—8)mRNA的表达。结果 (1)TGF—β1在正常组有少量表达 ,在JWYY组较正常组、模型组及假手术组显著升高 ,与法莫替丁 (FMTD)预防组比较差异有显著性。 (2 )IL—8在正常组几乎没有表达 ;模型组表达较正常组、假手术组、JWYY组和FMTD组显著增高 ;JWYY组和FMTD组比较 ,差别有显著性。结论JWYY能增加大鼠胃粘膜中TGF—β1mRNA的表达 ,减少IL—8mRNA的表达 ;对大鼠胃粘膜组织具有保护作用 ,预防乙酸诱导性胃溃疡。Objective To investigate the protecting effects of Jianwei Yuyang Granule (JWYYG) on gasric mucosa of rats with gastric ulcer and its mechanism responsible for this effect.Methods The model of gastric ulcer was prepared among SD rats by the modified Okabe method at first.Then,the expression of IL-8 mRNA and TGF-β1 mRNA was determined and compared by semi-quantitative RT-PCR among the rats of different groups treated with different therapies.Results The expression of TGF-β1 mRNA was not very significant among the rats of healthy group and was sighificantly increased among those of treating group with JWYYG,when compared with that of modeling group,sham-operated one and preventing one with famotidine(FMTD). On the other hand,the expression of IL-8 mRNA was at an extremely lower level among the rats of healthy group and was significantly higher among those of modeling group than ones in sham-operated group, JWYYG treated one and the preventing one with FMTD.Furthermore, significant difference could also be seen between that of JWYYG group and FMTD group.Conclusions JWYYG can give protecting effect on the gastric mucosa of rats to prevent suffering from gastric ulcer induced by acetic acid.This may be achieved at by increasing the expression of TGF-β1 mRNA and reducing that of IL-8 mRNA.
