产生西索米星的棘孢小单孢阻断型突变株JIM-121的筛选和研究

摘　　要：利用紫外光、氯化锂和甲胺复合因子处理棘孢小单孢JIM-202变株,得到一株JIM-121;其发酵液用离子交换树脂提取、然后用CM-Sephadex C-25分离表明主要含有二个抗生素即121-A和121-B。经薄层层析、红外光谱和核磁共振谱、质谱测定证实抗生素121-A与西索米星同质。根据庆大霉素-西索米星生物合成途径可推断该突变株系在西索米星到庆大霉素C_(1a)这一步转化绛红糖胺部份4′、5′还原反应较弱所致。The mutant JIM-121 was isolated after treating M. echinosPora strain JIM-202 with the complex effects of UV, LiC1 and methylamine. Antibiotic 121 complex was obtained from fermentation broth of JIM-121 with an ion-exchange procedure. By seperating and purifying with CM-sephadex C-25,antibiotic 121-A and 121-B were obtained as major components from antibiotic 121complex. Results of detection with TLC, IR, NMR and MS indicate that antibiotic 121-A is identical with Sisomicin. According to the biosynthestic pathway of Gentamicin and Sisomicin, we consider that the mutant strain,JIM-121,has the character of weak reducing ability at (4', 5')-position of purpurosamine.
