摘  要:目的:建立一种测定人尿液中血管抑素的方法,并探讨其在肿瘤患者中测定的意义。方法:依据ELISA的原理和技术要求,以赖氨酸作固相包被物,血管抑素为标准品,抗入K1~3的单抗为特异性的一抗,制备测定血管抑素的ELISA试剂盒。用此方法测定40例健康人(正常对照组)、92例恶性肿瘤(31例肺癌、31例食管癌、30例乳癌)患者治疗前尿中血管抑素含量。结果:该ELISA方法检测人尿中血管抑素的检出范围为1~200μg/L。正常对照组中尿血管抑素含量为0~27.4μg/L,x±s(16.15±6.82)μg/L。3种肿瘤患者尿中血管抑素平均水平均高于正常对照组(P<0.05),肺癌、食管癌、乳癌的诊断符合率分别为77.4%、71.0%和70.0%,其间差异无统计学意义(P>0.05)。结论:该方法操作方便,敏感性、特异性高;人尿中血管抑素含量检测可用于某些肿瘤的检出。Aim: To establish a method to detect Angiostatin levels in human arine and to explore the significance of the detection in cancers. Methods: According to the principle of ELISA technique, an assay for Angiostatin detection was established, including the 96-well plates coated with lysine, the purified Angiostatin as standard and the monoclonal antibody against human K1-3 as the first antibody. Angiostatin levels of urine collected from 40 healthy individuals and 92 cancer patents, including 31 cases of lung cancer, 31 cases of esophageal cancer and 31 cases of breast cancer, were measured by ELISA mentioned above. Results: The detection range was 1 - 200 μg/L. The concentration of Angiostatin in health group was 0 - 27. 4(16. 15±6. 82)μg/L. Tumor patients had a higher level of Angiostatin in urine than health group( P <0. 05). Patients with lung, esophageal and breast cancers had a correspondence ratio of 77.4% , 71.0% and 70.0% , respectively. Conclusion; The ELISA method mentioned above can be used to the Angiostatin level in human u-rine and to detect certain cancers.
