摘 要:本文采用反相高效液相色谱法测定人体血清及尿液中氟嗪酸的浓度。7名肾衰病人单剂量口服氟嗪酸300mg后药时曲线呈二室模型,平均达峰时间(t_(max))1.97±0.82h,药时曲线下面积(AUC)103.32±17.54mg·h/l,消除半衰期(t_(1/2)β)23.96±4.32h,其中6例病人氟嗪酸24h尿中排出给药量的18.34±7.31%,与正常人相比,肾衰病人的消除半衰期显著延长,尿排泄率明显降低。A RP-HPLC method has been used for the determination of ofloxacin in human serum and urine.The pharmacokinetics of ofloxacin was studied after a single oral dose of 300mg in 7 patients with renal failure. Serumconcentration-time data were fitted to a two-compartment model. The mean elimination t1/2β was 23.96±4.32 h, t_(max)1.97±0.82h, AUC 103.32±17.54 mg·h/l, respectively. The mean urinary recovery rate of ofloxacin over 24hafter administration was 18.34±7.31%. Compared with the data in normal volunteers, the elimination t1/2β was in-creased and urinary recovery rate was reduced obviously in patients with renal failure.
