养阴清肺方治疗肺癌的机理研究

摘　　要：目的　利用人肺鳞移植瘤模型研究平肺口服液的抗瘤作用机理。方法　采用人肺鳞裸鼠移植模型 ,分组分别给予生理盐水、平肺口服液及顺铂。给药 2 0d后无菌取瘤组织 ,观察肿瘤大小、肿瘤细胞增殖状况、肿瘤血管密度、细胞内皮生长因子 (VEGF)的蛋白表达。结果　平肺口服液能明显抑制肿瘤细胞增殖 ,但不及化疗组 ,化疗组肿瘤血管和模型组比有所减少 ,但不如平肺组减少明显 ,化疗组血管密度高于平肺组 ,平肺组的阳性细胞面积和平均视觉密度都明显少于模型组 ,化疗组VEGF阳性细胞染色深度有所增加。结论　以养阴清肺法为治则的平肺口服液对肺癌有明显的治疗作用 ,本研究初步证实抑制细胞增殖和影响肿瘤新生血管形成是其作用机理之一。Objective To study the anti tumor mechanism of the effects of Pingfei Oral Solution (POS) in the treatment of transplanted human pulmonary squamous cell carcinoma. Method The nude mouse model of transplanted human pulmonary squamous cell carcinoma was used in the experiment. The model mice were randomly divided into 3 groups, namely the normal saline group, POS group and cisplatinum group. The tumor tissue was isolated from the mice by aseptic manipulation 20 days after administration to observe the size of tumor, the proliferation of the tumor cells, the tumor vascular density, and the expression of VEGF. Results POS had remarkable effect of inhibiting the proliferation of carcinoma cells, but the effect was not as potent as that of cisplatinum. Cisplatinum could reduce the number of carcinoma vessels, but the effect was not as obvious as that of POS. The carcinoma vascular density was found to be higher in the cisplatinum group than that in the POS group. The area and the mean visual density of VEGF positive cells in the POS group were markedly decreased, as compared with those in the control group, while the intensity of the staining with respect to VEGF positive cells in the cisplatinum group was increased to a certain degree. Conclusion The POS representing the Yangyinqingfei therapy has a marked curative effect against transplanted human pulmonary squamous cell carcinoma. The results of this research initially prove that one of the mechanisms of the anti tumor effects of POS is inhibiting the proliferation of carcinoma cells and interfering neoplastic angiopoiesis.
