多巴胺诱导PC12细胞凋亡与多巴胺黑素生成的关系

摘　　要：目的　探讨多巴胺黑素 (dopamine melanin ,DA M )的生成在多巴胺 (dopamine ,DA)诱导神经元凋亡中的意义。方法　以PC12细胞为多巴胺能神经元的细胞模型 ,采用透射电镜和流式细胞仪 (flowcytometry ,FCM )观察细胞凋亡 ,紫外 -可见光分光光度计检测DA M。结果　DA可诱导PC12细胞凋亡 ,DA M的生成与DA诱导的PC12细胞凋亡之间呈正相关关系。结论　DAAIMTo study the significance of the formation of dopam ine-melanin (DA-M) in the apoptosis of PC12 cells induced by dopamine (DA). METHODSUsing PC12 cells as the model of dopaminergic neuron, DA-i nduced apoptosis in PC12 cells were observed by electron microscope and flow cyt ometry(FCM), and the formation of DA-M was detected by UV/VIS spectrometer. RESULTSDA induced PC12 cells apoptosis and there were positive corr elation between formation of DA-M and DA-induced apoptosis in PC12 cells. CONCLUSIONSThe formation of DA-M might play an important role in th e apoptosis induced by DA.
