早期自然流产与转化生长因子-β_1的相关性研究

摘　　要：目的　探讨自然流产患者外周血单个核细胞内转化生长因子 -β1(TGF-β1) m RNA的异常表达与自然流产的相关性。　方法　采用半定量逆转录 -聚合酶链反应 (RT-PCR)技术 ,检测3 0例早期自然流产患者、 2 5例正常妊娠妇女、 2 5例正常未妊娠妇女外周血单个核细胞内 TGF-β1m RNA表达量。　结果　早期自然流产患者、正常妊娠妇女、正常未妊娠妇女 3组研究对象外周血单个核细胞表达 TGF-β1m RNA相对含量分别为 (2 1.5± 9.4) %、(95.7± 12 .9) %、(87.0± 13 .8) % ,组间比较差异均有显著性 (P<0 .0 5)。　结论　正常早期妊娠妇女外周血单个核细胞 TGF-β1m RNA表达水平较正常未妊娠妇女显著升高 ,早期自然流产患者则显著下降 ,提示 TGF-β1对妊娠维护起重要作用 。Objective To investigate the expression of aberrant transforming growth factor beta mRNA by peripheral blood mononuclear cells(PBMCs) during early spontaneous abortion. Methods The expressions of transforming growth factor beta mRNA by PBMCs from 30 early spontaneous abortions,25 normal early pregnant women as well as 25 normal non pregnant women were measured by semi quatitative transcriptional polymerase chain reaction(RT PCR) method. Results we observed the levels of TGF β 1 mRNA from the above three groups which were(21 5±9 4) %?(95 7±12 9) % and (87 0±13 8) % respectively. Conclusion The level of TGF β 1 mRNA expression by PBMCs from early spontaneous aborters is significantly lower compared to those from the other two groups,whereas the level from normal early pregnant women is significantly higher.We propose that TGF β 1 may contribute to the maintenance of pregnancy,and abnormal expression of TGF β 1 may be associated with pregnancy failure.
