白松片对应激大鼠行为及额叶皮质5-羟色胺、多巴胺的影响

摘　　要：目的探讨中药白松片对应激大鼠行为及额叶皮质 5 羟色胺 ( 5 HT)、多巴胺 (DA)含量的影响。方法将健康成年雄性SD大鼠 60只 ,随机分为 :正常对照、模型对照、氟西汀对照组及白松片 3个剂量等 6组。通过强迫游泳建立应激大鼠模型。采用预防加治疗给药 ,观察开野试验得分、糖水消耗量、体重变化 ;高效液相 电化学检测法检测额叶皮质DA、5 HT含量。结果白松片各试验组开野试验水平得分 ( 71.0 0± 2 9.81～ 88.13± 2 4.74) ,垂直得分 ( 19.75± 8.63～ 2 6.75± 7.78)、蔗糖水消耗量、体重增加及额叶皮质 5 HT( 4 .2 1± 1.45～ 4.95± 1.41)、DA( 1.70± 0 .47～ 1.80± 0 .47)含量均高于模型对照组 (P <0 .0 5或P <0 .0 1)。结论白松片可改善应激大鼠抑郁行为 ,提高额叶皮质 5 HT、DA含量。Objective To explore the effects of Baisong-Tablet on the behavior and the levels of serotonin and dopamine in prefrontal cortex of stress rats. Methods60 adult Sprague-Dawley male rats were randomly divided into six groups including 3 control groups (Normal control group,Model control group and Fluoxetine hydrochloride control group) and 3 treatment groups (different dose of Baisong-Tablet treatment groups). The stress model was established by forced swimming . The changes of behavior were observed by open-field test(OFT) and the volume of sugar-solution drinked in 24 hours. Meanwhile, the weight changes were be observed.The levels of serotonin and Dopamine were measured by high performance liquid chromatography coupled with electrochemical detection. Results The scores of OFT and the volume of sugar-solution and the weight changes of Baisong-Tablet treatment groups were significantly higher than that of the model control groups, so were the levels of serotonin(4.21±1.45～4.95±1.41) and dopamine(1.70±0.47～1.80±0.47). ConclusionBaiSong-Tablet can reverse the depressive behavior by increasing the levels of serotonin and dopamine in cortex of stress rats.
