加味补肾壮筋汤颗粒剂防治去卵巢大鼠骨质疏松症的实验研究

摘　　要：目的 :评价与比较加味补肾壮筋汤传统汤剂与其中药配方颗粒剂对卵巢切除诱发骨质疏松大鼠模型的防治效果。方法 :选用 6月龄清洁级 Wistar雌性大鼠 60只 ,随机分为正常对照组。模型组、汤剂组和颗粒剂组 ,每组 1 5只。用双侧卵巢切除术进行动物造模 ,术后 1周给药 ,用药 3月后分别进行血清 Ca、P、ALP、BGP、PTH、CT、尿 HOP含量的测定 ,并进行全身骨、股骨颈、胫骨干骺端、腰椎等部位的骨密度检测。结果 :加味补肾壮筋汤汤剂和其颗粒剂均能明显增加血清 BGP、CT的含量 ,降低血清 PTH和尿 HOP含量 ,均能增加全身骨、股骨颈、胫骨干骺端、腰椎等部位的骨密度值 ,且差异与模型组有统计学意义 ( P<0 .0 5 ) ,但两者之间差异无统计学意义 ( P>0 .0 5 )。结论 :加味补肾壮筋汤颗粒剂进行剂型改革后仍能发挥防治骨质疏松症的作用 ,且与传统汤剂相比并不会导致疗效上的不同 ,故该中药配方颗粒剂可以代替传统汤剂使用。Objective:To evaluate the effect and difference of traditional decoction and formula granule of Jia Wei Bu Shen Zhuang Jin Tang on osteoporosis in ovariectomied rats.Methods:60 female Wistar rats aged 6 months were divided into four groups randomly:control group,model group,Traditional decoction group and formula granule group.Excising the ovaries of rats formed the animal's model of osteoporosis.All the rats were given drugs from one week after operation.3 months after experiment,the levels of calcium,phosphorus,alkaline phosphates (ALP),osteocalcin (BGP),parathyroid hormone (PTH),thyrocalicition (CT) in serum and hydroxyproline (HOP) in urine were measured.The bone mineral density (BMD) of the whole body skeleton,femoral neck,proximal tibia metaphases and lumbar vertebrae were detected simultaneously.Results:Both traditional decoction group and formula granule group were evidently increased the levels of serum BGP and CT,remarkably decreased the levels of PTH in serum and HOP in urine,Two treatment groups were both increased the bone mineral densities of the whole body skeleton,femoral neck,proxinal tibia metaphases and lumbar vertebrae.Two treatment groyps were significant difference to model group (P<0.05),otherwise insignificant each other (P>0.05).Conclusion:Not only the formula granule of Jia Wei Bu Shen Zhuang Jin Tang can be preserved osteoporosis in ovariectomied rats,but also its effect wasn't different to traditional decoction.So the formula granule of Jia Wei Bu Shen Zhuang Jin Tang may be widely used in- stead of traditional decoction.
