雌性大鼠成骨细胞内雌激素受体表达的研究

摘　　要：目的　研究雌性大鼠一生中成骨细胞内雌激素受体 (ER)表达水平的变化规律。方法　采用二次酶消化、反复贴壁法分离纯化培养大鼠成骨细胞并进行鉴定 ,应用半定量Westernblot对不同月龄雌性Wistar大鼠成骨细胞内雌激素受体进行检测。结果　ER表达在 1、2月龄组呈低度表达 ,3月组ER开始呈现上升趋势 ,至 8、9、1 0月组ER达到高峰 ,1 0月组以后表达呈现下降趋势。结论　ER表达在不同月龄大鼠呈规律性分布 。Objective To detect estrogen receptor in osteoblast and its regulation of expression. Methods The osteoblasts were isolated from different months female Wistar rats long bones by sequential enzyme digestion, partial osteoblasts were cultured by DMEM, and osteoblast was detected.Results The ER indicated by Western blot showed low expression in rats of 1 month, rats of 2 month groups, which showed an upward trend in 3 month groups. The ER indicated by Western blot showed the most intensive in 8, 9, 10 month groups, and then descended beyond 10 month groups.Conclusion The ER expression is in the regular distribution in different months. The estrogen receptor expression is in connect with the level of estrogen.
