摘  要:目的 观察纤粘连蛋白 (fibronectin ,FN)诱导的Hela细胞的黏附及基质金属蛋白酶 (matrixmetalloprotein ases,MMPs)基因的表达 ,探讨恶性肿瘤细胞侵袭、转移发生的机制。方法 无血清培养Hela细胞 ,以FN作为细胞外基质 (extracellularmatrix ,ECM)成分 ,用MTT法检测Hela细胞的黏附率及光镜下计数Hela细胞的铺展率并观察Hela细胞形态的变化 ;用明胶酶谱法检测MMPs的活性。结果 无FN作用的Hela细胞不发生黏附及铺展 ,亦无明显MMPs基因表达 ;当纤粘连蛋白浓度为 5~ 1 0 μg ml时 ,黏附率及铺展率最大 ;黏附的Hela细胞启动MMP 2基因的表达 ,纤粘连蛋白浓度为 5~ 1 0 μg ml时 ,MMP 2活性最大。 结论 FN可诱导Hela细胞黏附 ,进而引发一系列细胞内信号转导机制 ,启动MMP 2基因表达 。Objective To investigate the adhesion of Hela cell and the expression of MMP 2 induced by fibronectin(FN),to study the mechanism of the invasion and the transfer of tumor.Methods Hela cell was cultured in RPIM1640 without calf serum and in FN as extracellular matrix.MTT was used for the detection of the adhesive rate of Hela cell,counted the spread rate and observed the structure of Hela cell under the inverted microscope; gelatin zymography was used for the detection of the activity of MMPs.Results Without FN,there was no adhesion and spread of Hela cell,and no expression of MMP 2 gene; when the concentration of FN reached 5~10μg/ml the adhesive rate and the spread rate was the highest; when the concentration of FN reached 10μg/ml after 12h ,the activity of MMP 2 was the highest.Conclusion Hela cell can be induced by FN to adhere which causes a series of signal transductions in cell and expresses MMP 2 gene,accelerates the transfer of tumor.
