摘  要:目的 采用 5 氮杂胞苷 (5 aza)诱导骨髓间充质干细胞分化成心肌细胞。方法 分离培养骨髓间充质干细胞 ,采用密度为 1 .0 73的percoll分离液分离大鼠骨髓间充质干细胞并进行体外培养和连续传代 ,在第 3代细胞以5 氮杂胞苷进行诱导 ,用免疫组化的方法鉴定心肌特异性肌钙蛋白I的表达。结果 诱导前 ,骨髓间充质干细胞形态均一 ,呈多突起扁平形态 ,诱导后细胞形态发生变化 ,细胞之间形成连接 ,排列方向渐趋一致 ,免疫组化显示表达阳性。结论  5Objective To study 5 azacytidine (5 aza) which could induce differentiating of mesenchymal stem cells(MSCs) into cardiac myocyte.Methods MSCs were isolated from adult rats. The MSCs were purified by culturing, expanding and expanding. The third passage MSCs were treated by demethylating agent 5 azacytidine. The expression of cardiac Troponin I (cTnI) was observed by immunohistochemistry.Results After the MSCs were treated with 5 aza, the shape of cell changed. The connections of the cells were formed. The direction of the cell arraying was similar gradually. Immunohistochemistry showed that the expression of cardiac Troponin was positive. Electron microscope results showed that fusiform cell had apparent myofilament. The central of cell had an oval shap nucleus.Conclusion 5 aza might induce MSCs differentiating into cardiac myocyte.
