摘 要:目的 观察脑室注射孤啡肽 (OFQ)和吗啡对大鼠皮层体感区Ca2 + ATP酶活性的影响。方法 检测Wistar大鼠皮层体感区组织匀浆上清液Ca2 + ATP酶活性。观察①脑室注射吗啡对大鼠皮层体感区Ca2 + ATP酶活性的影响 ;②脑室注射OFQ对大鼠皮层体感区Ca2 + ATP酶活性的影响 ;③两侧脑室同时注射吗啡和OFQ对大鼠皮层体感区Ca2 + ATP酶活性的影响。结果 ①脑室注射 2 0 μg 2 0 μl吗啡 6 0min后 ,大鼠皮层体感区Ca2 + ATP酶的活性与生理盐水组相比明显降低 (P <0 .0 1 )。②脑室分别注射不同浓度的孤啡肽 (0 .0 4、0 .4 5、0 .9、1 .8μg 2 0 μl) 6 0min后 ,皮层Ca2 + ATP酶活性随OFQ浓度的增加而降低 ,组间差异显著 (P <0 .0 5 )。③右侧脑室注射 2 0 μg 2 0 μl吗啡 ,同时左侧脑室注射 0 .9μg 2 0 μl孤啡肽 6 0min后 ,可相互拮抗对Ca2 + ATP酶活性的影响。结论 脑室单独注射吗啡或孤啡肽均可使大鼠皮层Ca2 + ATP酶的活性降低 ,而孤啡肽 (0 .9μg 2 0 μl)又可以拮抗吗啡对Ca2 + ATP酶活性的抑制作用 ,说明孤啡肽在中枢通过影响Ca2 + ATP酶活性产生抗阿片作用 ,从而影响痛觉调制。Objective To investigate the effect of intracerebroventricular injection of morphine and orphanin FQ(OFQ) on Ca 2+ ATPase activity of rat cerebral cortex.Methods The cerebral cortex tissues were extracted, homogenized and centrifuged. Ca 2+ ATPase activity was measured. To investigate: ①Effect of intracerebroventricular injection of morphine on Ca 2+ ATPase activity of somatosensory area of rat cerebral cortex; ②Effect of intracerebroventricular injection of OFQ on Ca 2+ ATPase activity of somatosensory area of rat cerebral cortex; ③After intracerebroventricular injection of morphine at right and OFQ at left at the same time, the change of Ca 2+ ATPase activity of somatosensory area of rat cerebral cortex.Results ①After intracerebroventricular injection of morphine (20μg/20μl), the Ca 2+ ATPase activity was significantly decreased as compared with control group ( P <0.01). ②After intracerebroventricular injection of OFQ (0.04, 0.45, 0.9, 1.8 μg/20μl), the result was that the Ca 2+ ATPase activity was significantly decreased with the increase of concentration of OFQ, and the difference among the groups were significant ( P <0.05). ③After intracerebroventricular injection of morphine at right and OFQ at left at the same time, the Ca 2+ ATPase activity was not significantly different as compared with control group.Conclusion These results indicate that morphine or OFQ can reduce the activity of Ca 2+ ATPase, while OFQ and morphine can antagonize the inhibiting effect of Ca 2+ ATPase. It suggest that the effect of OFQ is related to the changes of Ca 2+ ATPase activity, and OFQ can reverse the effect of morphine in CNS.
