丹仙康骨胶囊对培养成骨细胞影响的观察

摘　　要：目的 :为了解补肾活血中药丹仙康骨胶囊对体外培养成骨细胞的作用。方法 :应用透射电镜、MTT、对硝基苯磷酸盐法 (PNPP)及骨钙素 (BGP)含量放免测定法 ,观察丹仙康骨胶囊对成骨细胞超微结构、增殖与分化作用的影响。结果 :丹仙康骨胶囊刺激 (2 0mg/ml)的成骨细胞 ,ALP活性提高及骨钙素含量增多 ;透射电镜观察其细胞线粒体致密、游离核糖体增多、内质网丰富扩大增粗呈中等电子密度 ,而糖原溶解与脂肪空泡均较少。结论 :丹仙康骨胶囊具有促进成骨细胞的代谢。Objective:To investigate the effects of Danxian Kanggu(DXKG) capsule extracted from traditional Chinese medicine on osteoblast cultured in vitro.Methods:Transmission electron microscope,MTT,PNPP and BGP content radioimmunoassay were used to observe the effect of DXKG capsule on the ultrastracture proliferation differentiation of osteoblasts.The activity of ALPase and the BGP content of osteoblasts cultured in vitro.Results:DXKG capsule had the effects on stimulated osteoblasts that ALPase activity rised and BGP content increased in different concentration,expecially in 20 mg/ml concentration.Under transmission electron microscope observation,changs could be seen in stimulated osteoblasts by DXKG such as the mitochondria were more fine and close and free ribosome were increased and the endoplasmic reticulum were more rich and large,showed middle electron density than that of the control group,but glycogen dissolution and liposome vacuoles obviously decreased.Conclusion:DXKG capsule possesses effects on stimulating proliferation and differentiation such as increasing the activity of ALPase and the BGP content of osteoblasts cultured in vitro.
