摘  要:目的 :研究人参总皂甙 (TSPG)对人淋巴细胞表达早期造血生长因子的影响 ,探讨人参“补气生血”的现代分子生物学机理。方法 :采用造血祖细胞体外培养、造血生长因子生物活性检测、免疫细胞化学、核酸分子原位杂交技术 ,研究TSPG对胎儿胸腺细胞、脾细胞表达早期造血生长因子的影响及其机理。结果 :经TSPG诱导制备的胎儿胸腺细胞条件培养液 (HTCM )、脾细胞条件培养液 (HSCM)对人髓系多向性造血祖细胞 (CFU Mix)的增殖分化有明显促进作用 ;经TSPG诱导后胎儿胸腺细胞、脾细胞内IL 3的蛋白及mRNA表达显著提高。结论 :TSPG可能诱导人淋巴细胞表达早期造血生长因子 ,从而促进人早期血细胞增殖分化。Objective:To study the effect of total saponins of Panax ginseng (TSPG) on expression of early hematopoietic growth factor in human lymphoclytes, and to explore the cellular and molecular biologic mechanisms of Panax ginseng 'invigorating qi and hematopoiesis'. Methods: The techniques of culturing hematopoietic progenitor cells in vitro, bioassay of hematopoietic growth factor (HGF), immunocytochemistry and nucleic acid in situ hybridization were used to study the effect of TSPG on early hematopoietic growth factor expression in fetus thymocytes and splenocytes. Results: The conditioned culture media of thymocytes and splenocytes prepared with TSPG promoted the proliferation and differentiation of CFU-Mix; the protein and mRNA expression of IL-3 in thymocytes and splenocytes induced by TSPG was greatly intensified. Conclusion: TSPG may induce human lymphocyte to express early hematopoietic growth factor, thus promote the proliferation and differentiation of human hematopoiesis at early stage.
