摘 要:目的 探讨阿霉素对肾损伤的毒性机制。方法 用治疗剂量阿霉素静脉注射,建立兔阿霉素肾损伤模型作为模型组,静脉注射生理盐水作为对照组。取肾组织进行HE梁色,采用Gomori法、Padykula及Herman法和NADPH-d酶组织化学染色;进行图像发析,数据进行统计学检验;观察和比较两组动物肾组织中ALP、Ca^(2+)-ATP酶和NOS酶的变化。结果 病理学检查表明模型组肾组织受损伤,酶组织化学染色和图像分析显示ALP、Ca^(2+)-ATP酶含量下降,而NOS表达上调,模型组与对照组相比,P<0.01,具有显著性差异。结论 治疗剂量阿霉素能够降低肾组织中ALP、Ca^(2+)ATP酶的活性和上调NOS的表达,可能是导致肾损伤的重要原因。Objective To explore the toxic mechanism of adriamycin (Adr) in the kidneys damaged in rabbits. Method The therapeutic dosage of Adr was injected into vein of rabbits in model group and saline was injected in control group. HE staining and enzyme histo-chemical methods were used and image analysis was performed, ALP.Ca2+-ATPase and NOS in kidneys of two groups were observed and compared. Result Pathological examination showed the kidneys damage. Enzyme histochemical examination showed ALP.Ca2+-ATPase activities were reduced and expression of NOS was increased in model group,P< 0.01. Conclusion The therapeutic dosage of Adr reduced ALP,Ca2+-ATPase ctivities and increased the expression of NOS, Which may cause Kidney damage.
