摘  要:采用含不同成分的MS培养基处理立碗藓的原丝体 ,发现随蔗糖浓度的提高原丝体的生长相对趋于老化 ;当培养基中附加不同浓度的 2 ,4 D和 6 BA时 ,1 0日龄的原丝体出现了明显变化 ,当二者浓度均为 0 .5mg·L 1 ,幼嫩的原丝体上能长出疏松易碎的白绿色愈伤组织。利用 1 %纤维素酶和 0 .8%的果胶酶分别处理 1 0日龄、2 0日龄的原丝体和 1 0日龄的茎叶体的叶片 2~ 1 0h ,结果发现1 0日龄的原丝体在酶解 2h后能游离出大量的原生质体 ;而 2 0日龄的原丝体效果较差 ,只能游离出极少量的原生质体 ;对于 1 0日龄的茎叶体酶解 1 0h仍看不到原生质体游离出来。这表明立碗藓原丝体对不同的外界条件可产生不同的反应 。The protonema of Physcomitrium sphaeri was treated by the MS media supplemented with different ingredients, the result showed that the protonema got older with increasing the concentration of sucrose; And there were big changes when the 10-days old protonema was cultured on the media with different concentration of 2,4-D and 6-BA, the white-green crimple callus could be induced on MS media supplemented with 2,4-D 0.5 mg·L -1 and 6-BA 0.5 mg·L -1. When the protonema of 10-days old and 20-days old and 10-days old leaves were digested by 1% Cellulase and Pectolyase Y-23 respectively, we found that, there were large amount of protoplasts isolated from 10-days old protonema two hours later; But to 20-days protonema, only few of them could be isolated from the material; For the leaves derived from 10-days old cauliform, we couldn't see any protoplasts after digesting 10 hours. These results showed that the protonema of Physcomitrium sphaeri could react on different conditions, but its state was very important to the reaction.
