摘  要:目的 :观察丹参预防自发性高血压大鼠 (SHR)左室肥厚 (L VH)的作用 ,并观察其对心肌血管紧张素 受体 1(AT1R)表达的影响。方法 :18只 8周龄的 SHR大鼠随机分成 3组 ,每组 6只。S8组于 8周处死 ,另两组分别经腹腔按 1g· kg- 1 · d- 1 剂量注射丹参 (D18组 )或蒸馏水 (S18组 ) ,共 10周。测量大鼠尾动脉收缩压 (SBP)及左心室质量指数 (L VMI)。应用苏木素伊红 (HE)和 Van Gieson(VG)染色、免疫组织化学方法 ,并结合计算机图像分析技术 ,检测心肌细胞的直径 (TDM)和面积 (CA )、心肌组织胶原体积分数 (CVF)、血管周围胶原面积与管腔面积比例 (PVCA) ,以及 AT1R的表达。结果 :与 8周龄的 SHR大鼠相比 ,18周龄 SHR大鼠的 SBP、L VMI、TDM、CA、CVF和 PVCA均显著增加 ,AT1R表达明显 ;丹参治疗可抑制 L VH的发展和心肌组织 AT1R的表达 ,但对 SBP无明显改变。结论 :长期应用丹参治疗可预防 SHR大鼠 L VH的形成 ,其机制可能与丹参降低了心肌细胞 AT1R的表达有关。Objective: To observe the effect of salvia miltiorrhiza bge(SMB,丹参) on preventing left ventricular hypertrophy(LVH) in spontaneously hypertensive rats(SHR) and the expression of angiotensin Ⅱ receptor type 1(AT1R). Methods: Eighteen SHR rats of 8-week-old were randomly divided into three groups(n=6). The rats in S8 group was killed at 8 weeks. SMB or distilled water(1 g·kg -1·d -1) was injected intraperitoneally into the other two groups for 10 weeks. Systolic blood pressure(SBP) and left ventricular mass index(LVMI)were measured. Hematoxylin and eosin(HE), Van Gieson(VG) and immunohistochemical staining combined with computed morphometry were imployed to evaluated the cardiomyocyte area(CA) and transverse diameter of myocardial cells (TDM), the collagen volume fraction (CVF), perivascular collagen area/vascular lumenal cross area(PVCA) and AT1R expression in the left ventricular tissue. Results: Compared with 8-week-old rats, the SBP, LVMI, TDM, CA, CVF, PVCA and AT1R expression increased markedly in the 18 th week of SHR rats. Treated with SMB, the LVH was significantly prevented and AT1R expression decreased whereas SBP changed slightly. Conclusion: Chronic treatment with SMB can inhibit the development of LVH in SHR rats, which is probably related to the decease of cardiac AT1R.
