银杏提取物对淀粉样β蛋白致PC12细胞凋亡的保护作用

摘　　要：目的:探讨银杏提取物对淀粉样β蛋白(Aβ)致PC12细胞凋亡的保护作用。方法:应用二甲基噻唑二苯基四唑溴盐(MTT)分析法研究细胞活性的改变,应用激光共聚焦显微镜进行线粒体膜电位测定。TUNEL法测定细胞凋亡率。结果:随着Aβ浓度的加大,PC12细胞的存活率逐渐降低。同时给予Aβ及不同剂量的银杏提取物,可明显提高PC12细胞的存活率。银杏提取物可减少Aβ所致的细胞凋亡,Aβ组细胞凋亡率为63%,高于银杏组21%(χ2=36.2069,P=0.0000)。结论:银杏提取物可抑制Aβ诱导的PC12细胞凋亡。AIM:To investigate the effect of ginkgo biloba extract(GBE) in protecting PC12 cells from apoptosis induced by amyloid beta protein(Aβ). METHODS:Survival rate of PC12 cells was detected by MTT method. Mitochondrion membrane potential(MMP) was tested by laser confocal microscopy.The rate of cell apoptosis was detected by TdT mediated biotinylated dUTP nick end labeling(TUNEL) method. RESULTS:With the increase of Aβconcentration,cell survival rate was decreased gradually.However,the survival rate of PC12 cells were significantly increased following the treatment of Aβand different dosage of GBE simultaneously.MMP damage and cell apoptosis induced by Aβcan be ameliorated by the treatment of GBE.The apoptotic rate was 63%for the Aβgroup, significantly higher than in the GBE group(21%)(χ2=36.206 9,P=0.000 0). CONCLUSION:GBE can inhibit PC12 cell apoptosis induced by Aβ.
