玻璃体内注射氯喹对兔视网膜电图和超微结构的影响

摘  要:本文采用玻璃体内给药法,以暗适应视网膜电图(ERG)为主要指标,结合视网膜超微结构的变化,观察了氟喹对兔视网膜的毒性作用,结果,ERGa,b波的抑制程度与注射剂量(0.3~3.0mg)和药后的时间呈现出相应的关系,超微结构显示,药后48h,视网膜的感光细胞和双极细胞的线粒体明显肿胀,感光细胞外段膜盆结构排列紊乱,氯喹玻璃体注射所引起的急性ERG变化,与临床慢性口服氯喹中毒相似。The present work was undertaken to evaluate the acute effects of intravitreous injection of chloroquine on ERG and ultrastructural changes in rabbits. Albino rabbits were sedated and the pupils were dilated. Chloroquine (0.3 to 3.0 mg·0.1 ml-1)and saline (0.1 ml) were injected into the vitreous body of the left eye and right eye (as control), respectively. Scotopic ERG was measured before and 4, 24, 48 h and 8 d after injections. The results were expressed as amplitude ratios of ERG-waves by comparing the treated eye with the untreated one for each animal. After injecting the drug amplitude ratios of the waves were declined as the doses were increased and the time was prolonged. Obvious dose- and time-dependent effects were exhibited. The reduction of the b-wave amplitude ratio happened prior to that of the a-wave amplitude ratio. 4 h after injection 1 mg and 3 mg drugs, the b-wave amplitude ratio, as compared with that before the injection, was decreased significantly, but the reduction of the a-wave amplitude ratio was not obvious in 4 h after injection. When the ERG was measured 48 h and 8 d after the injection, a trend towards the recovery of ERG appeared in the eyes treated with 1 mg chloroquine, while the ERG of eyes in-jected with 3 mg chloroquine disappeared completely. At 48 h after injecting 1 mg and 3 mg chloroquine, the electron-micrographs showed that the mitochondria of the photoreceptor and bipolar cells were swelling and degenerative. The disturbance of lamellar disk architecture was observed in photoreceptors. The present study shows that the immediate changes of the a- and b-wave after intravitreous injection of chloroquine indicate a direct neuroretinal toxicity.
