HPLC测定Aztreonam血药浓度及药代动力学的研究

摘　　要：本文建立了测定血清中 Aztreonam 浓度的高效液相色谱法,固定相:ODSC_(18),流动相:甲醇-磷酸盐缓冲液(1:9,v/v);检测器:UV 254nm;流速:1.0ml/min;保留时间:8min,标准曲线线性范围:2～200μg/ml,r=0.9999;平均回收率:100.95%±0.03%,在6只家兔药代动力学研究的基础上,研究5名健康志愿者药代动力学的特点,并用计算机拟合了药代动力学的参数,得半衰期(T_(1/2))平均为:1.2523±0.1898h,平均峰浓度:297.34±77.93μg/ml,平均表观容积为:4.1809±0.2474(L),并考察了5名患者的峰浓度和谷浓度(iv 1.0g/次,tid)。HPLC method was developed to determineserum concentration of aztreonam.Fixed phase wasODS C_(18),and mobile phase was methanol:phosphatebuffer=1:9(v/v).Detection was set at UV 254nm with a flow rate of 1.0ml/min.Retention timeof aztreonam was 8 minutes.Linear range was 2～200μg/ml,r=0.9999.The average recovery was100.95%±0.03%.Pharmacokinetic parametersof aztreonam in six rabbits and five healthy volun-teers were calculated by computer.Some parameterswere as follows:T(1/2)=1.25±0.19h,C_(max)=297.34±77.93μg/ml,and Vd=4.18±0.25L(mean±SD).Peak and trough level of aztreonamin five patients(iv 1.0g,tid) were measured.
