Behcet’s病患者外周血和结节性红斑组织中T-bet基因的表达及意义

摘　　要：目的观察Th1细胞特异性转录因子T-bet(T-boxexpressinTcells)在活动期Behcet’s病患者外周血单个核细胞和结节性红斑组织中的表达情况,探讨T-bet在Behcet’s病发病中的作用和地位。方法收集14例活动期Behcet’s病患者和12例正常人外周血,采用Western免疫印迹检测外周血单个核细胞的T-bet的表达水平;对4例Behcet’s病活动期患者腿或臂部结节性红斑组织进行免疫组织化学染色观察的T-bet表达。结果14例活动期Behcet’s病患者均可检测到相对分子质量约为62000的特征性免疫印迹带,所有的正常对照却没有发现。正常人皮肤表皮T-bet分子表达均为阴性,4例结节性红斑表皮T-bet分子均表达阳性,表皮阳性细胞数为108个±10个/mm2,阳性表达率为35%～42%。结论活动期Behcet’s病外周血单个核细胞和结节性红斑组织T-bet的表达均显著增高,T-bet分子可能在Behcet’s病发病机制中起重要作用。Objective To investi ga te the expression of Th1-specific T-box transcription factor T-bet in periph eral b1ood mononuclear cells(PBMCs)and erythema nodosum of Behcet’s disease (BD)patients with active intraocular inflammation,and analyze the correlation between the expression of T-bet and inflammation in BD.Methods Blood samples were taken from14BD patients with active intraocular inflammation and12heal thy individuals.PBMC were isolated by centrifugation with Ficoll-paque,and T -bet protein expression was assessed by Western blot analysis.SP immunohistoch emical method was used to detect the expression of T-bet in erythema nodosum f rom4patients with active Behcet’s disease and normal skin from2healthy indivi duals.Results A62kDa band consistent with T-bet by Western blot was detect able in BD patients with active uveitis,but no in controls.The negative stain ing of T-bet was found in the normal skin,but in the erythema nodosum,appro ximately35%～50%of expression rate in the epithelial cells with the positi ve number of l08±10.Conclusion Behcet’s disease is associated with an up -regulation of T-bet expression.T-bet positive lymphocyte cells may be respo nsible for the incidence and consistence of inflammation in erythema nodosum.
