摘 要:浮力密度相同的粒子,在区带转头里用等容积蔗糖梯度分离的效果,优于等速蔗糖梯度和线性蔗糖梯度。本文给出用于日立RPZ48T,RPZ35区带转头,实验参数为:温度5℃,20℃,初始蔗糖浓度5%(W/W),20%(W/W),粒子密度1.33(g/cm^3),1.41(g/cm^3),1.89(g/cm^3)条件下构成等容积蔗糖梯度所需的容积与蔗糖浓度的数值,及用日立DGP型密度梯度程序泵配制此梯度液的方法。The effectiveness of separation for particles with equal buoyant density by the method of equivolumetric sucrose gradient is higher than that of equal velocitysucrcse gradient with zonal rotors, Under conditions of 5℃, 20℃, initial concentrations of 5% (w/w) and 20% (w/w), sucrcse solution, particle density 1.33 (g/cm^3) and 1.89 (g/cm^3), respectivcly, the volume and the concentration of sucrose solution which are necessary in the method of equivolumetric sucrose gradient with the zonal rotor equipped on Hitachi RPZ48T, RPZ35 have been obtained and the way of preraration of the gradient solution made by pump (Hitachi DGP) programmed for density gradient has also been discussed in this paper.
