摘 要:目的 研究红景天对体外培养视网膜色素上皮细胞增生的抑制作用及可能机制。方法 通过活细胞计数法与MTT比色法分别检测不同质量浓度 ( 5 0 0、40 0、3 0 0、2 0 0、10 0、5 0、10 μg/mL)红景天和红景天 2 0 0 μg/mL在不同作用时间 ( 6~ 12 0h)对RPE细胞增生及代谢的影响。结果 红景天组细胞增生受抑制并出现细胞脱落 ,红景天 40 0 μg/mL具有最强的抑制效应 ,其明显抑制效应在 6h即出现 ,72h达高峰。红景天组A值明显低于对照组 ,RPE细胞生存率下降。结论 红景天可能通过干扰RPE细胞代谢对RPE细胞增殖具有剂量依赖和时间依赖方式的抑制作用。ObjectiveTo investigate the effects of Rhodiola rosea on proliferation of cultured retinal pigment epithelium(RPE) cells and the probable mechanisms.MethodsCultured RPE cells were treated by various concentrations of (500, 400, 300, 200, 100, 50, 10 μg/mL) Rhodiola rosea at different times(6-120 hours). Cells proliferation and metabolism were evaluated using cell count, MTT colorimetric assay and 3 H-TdR.ResultsRhodiola had inhibition on proliferation of RPE cells, and some cells detached from the wall. The strongest inhibitory concentration of Rhodiola rosea was 400 μg/mL. The significant inhibitory effect occurred at the 6th hour with the peak time at the 72nd hour. The A values in Rhodiola rosea groups were significantly lower than those in control group,and the survival rate of RPE cells was lowered. The inhibitory rate was enhanced with Rhodiola level increased.ConclusionRhodiola rosea could inhibit the proliferation of RPE cells in a dose-dependent and time-dependent manner.The probable mechanism is that Rhodiola rosea decrease the synthesis of DNA and disturbe the metabolism of RPE cell.
