摘 要:为研究apoA-Ⅳ功能,用密度梯度超速离心人血清获得LDS,经与Intralipid孵育及SDS-PAGE纯化得apoA-Ⅳ,鉴定为SDS-PAGE纯,分子量46,000.分析了我国人血清apoA-Ⅳ氨基酸组成及等电聚焦pI值(5.27和5.47),结果与国外报道基本一致。研究了apoA-Ⅳ及其脂质体(apoA-Ⅳ:DMPC)对人皮肤成纤维细胞胆固醇的外流作用,对照组与apoA-Ⅳ组无显著差异(P>0.5),而对照组与脂质体组有非常显著的差异(P<0.001),显示apoA-Ⅳ脂质体有逆向转运胆固醇的作用,这可能为动脉粥样硬化的防治提供一个新的途径。In order to study the function of apoA-Ⅳ, hunan serum apoA-Ⅳ was isolated and purified. LDS was isolated with NaBr density gradient ultracen-trif ugation and then incubated with intralipid according to the method of Wein-berg and Scanu. Protein complex containing apoA-Ⅳ was obtained. The apo-Ⅳ was purified from protein complex by preparative SDS-PAGE, elec-troelution and renoval of SDS. ApoA-Ⅳ thus prepared gave a single band on SDS-PAGE with a MW of 46 kD. Amino acid co nposition and isoelectric focusing pI (5.27 and 5.47) of apoA-Ⅳ were similar to those reported in the literature. The effects of apoA-Ⅳ and liposome (apoA-Ⅳ: DMPC) on cholesterol efflux from hunan skin fibroblast were studied. There was no significant difference (P>0.5) between apoA-Ⅳ and control but a very significant difference (P<0.001) between liposo.ne (apoA-Ⅳ: DMPC) and control, suggesting that liposomes (apoA-Ⅳ:DMPC) play a role in reverse cholesterol transport. The results may offer a new approach for the prevention and treatment of atherosclerotic coronary heart disease.
