摘 要:本文以石墨炉原子吸收分光光度法为测定方法,对国产卡铂和顺铂在家兔体内的药物动力学进行了初步的研究和比较。本法中卡铂和顺铂在0~20μg/ml范围内药物血浆浓度同吸收峰高呈线性关系,相关系数r>0.99(P<O.01),最低检测浓度≤0.2μg/ml,精密度的日内变异系数CV均低于5%,日间CV皆低于9%。单剂量一次静脉推注给药,卡铂剂量为6mg/Kg,顺铂剂量为5mg/Kg,结果,两药在兔体内的动力学过程符合线性开放二定模型。卡铂在体内的消除显著大于顺铂,两药的动力学参数,除中央室表观分布容积Vc外,其余各相应参数间均有显著或非常显著性差异。An initial study and comparision on pharmacokinetics of carboplatin and cisplatin native to China inrabbit body were presented by GAAS method. In the method, standard curves of carboplatin and cisplatinwere fitted to the linear equation: plasma concentration of drugs was linear with the height of absorptionpeak over the range of 0-20μg/ml, the correlation coefficient was about 0.99 (P<0.01), the limit of de-tected concentration was 0.2μg/ml. The variation of CV. was less than 5% within-day, and less than 9%between-day. Single dose was injected intravenously with 6mg/kg of carboplatin and 5mg/kg of cisplatinrespectly. The resulte showed that kinetics process of two drugs was fitted to linear two-apartment modelin rabbit body. The body elimination of carboplatin was higher than that of cisplatin. Besides central com-partment VC, all other parameters had more or most significant difference between the two drugs.
