高渗盐水对顺铂毒性及药物动力学的影响

摘  要:本文以血清尿素氮及体重做为毒性评价指标,高渗盐水减低了顺铂的毒性。小鼠静注15mg/kg顺铂后,血浆总铂的药物动力学呈三室模型。高渗盐水缩短了血浆总铂的α相半衰期,提高了给药后各时间点血浆超滤铂的浓度,降低了肾脏内给药后2h内的铂浓度,没有明显改变肿瘤内铂浓度的药时曲线下面积。实验表明,高渗盐水提高了顺铂的治疗指数。The cisplatin induced toxicity. indicated by the elevation in blood urea nitrogen concentration and body weight loss. couldbe reduced by hypertonie saline (HS) used as a vehicle of cisplatin. The influence of HS on pharmacokinetics of cisplatin inmice was analysed. The disapper ance of total plasma platinum was well described by a three compartment model. The α-phaseof total platinum was shortened in HS group. HS vehicle resulted in significant higher levels of ultrafiltrate platinum. In HSgroup, the kidney platinum concentration was reduced in the initial two hours after cisplatin administration. The platinum con-centration in tumor tissue of HS group was similar to the control group in the initial 40 min and was lower than the controlfrom 1 to 12 hour after cisplatin administration. The AUC? of platinum in tumor tissue in HS group was not decreased signi?icantly compared to the control. The results suggested that using HS as a vehicle of cisplatin may be benificial to patienttreated with cisplatin.
